Key on, engine off (no vacuum to regulator), 43.5 psi is the approx. design pressure. Engine running, mid to upper 30's psi. Manifold vacuum is applied to the pressure regulator with the engine running to keep the differential pressure across the injectors constant. If you blip the throttle with the engine running the fuel pressure should spike upwards momentarily and then settle back.
